I’m having trouble looking up the proper dosing for “smoking toad.” Do you straight out smoke it? Freebase? Throw it on the barbie? Because I’ve heard both terms referred to as smoking, and yet they would (probably) give very different onsets and duration, so would require different dosing regimens. Is one dose the same for most people, or should it be weight based and dose in mcg/kg?

And without knowing proper dosing, how can we know if the poor fellow overdosed?

This sensationalistic article is more worthy of the National Enquirer than the normally psychedelic-savvy BoingBoing. The term “venom” is misapplied here. 5-MeO-DMT is a quite powerful and valuable traditional South American psychedelic medicine. It is being used (discretely) to help people heal a wide variety of mental issues, including PTSD in veterans and other trauma. As others have pointed out above, we don’t know a thing about the cause of death, just that the alleged victim also at some point used the toad. I don’t know that 5-MeO even has a toxic dose and it seems unlikely it caused any death, let alone murder. Wikipedia doesn’t mention anything at all about any toxicity: 5-MeO-DMT - Wikipedia

Edit 1: Apparently, if you’re dumb enough to orally ingest a large amount of Sonoran Desert toad secretion it can kill you. Smoking pharmacologically-active doses probably not. Know your dose!
Edit 2. Wikipedia seems to be mistaken in citing it’s use by shamans in South America. (The article they cite is only an abstract and it’s behind a paywall.) It appears to be native to North America; it has been used ceremonially in Mexico and the Southern U.S.

Ayahuasca, which has a closely related DMT molecule has been used for a long time in South America, but Incilius alvarius is strictly North American. If they actually used the toad toxin directly it also contains bufotenin which has an LD50 of around 250 mg/kg (~20 g for an 80 kg person). There have been instances of people dieing of bufotenin overdose from eating toads, but I don’t think anyone who was knowingly ingesting the toxin. Unfortunately there is no useful info on the DMT source or how they ingested it, so who knows

Cane toad venom fatalities are pretty common. It’s a traditional Chinese medicine.
