I would use a comma if the two subjects were not paired. For example;
What did you have for lunch today? “Peanut butter,[comma] and sushi”, sounds like separate dishes. Whereas to my eye, “Peanut butter and sushi”, reads like some kind of fusion dish.
As always, your interpretation may vary.
